Network topology awareness (NTA) is a fundamental issue in the context of computer networks: the lack of control over the topology can negatively impact performance, security, resilience, and so on. However, in Mobile Ad-hoc NETworks (MANETs), NTA is difficult to achieve because of their dynamicity. Different reactive protocols for MANETs, in which routes are established only when needed, provide different NTA to each host, depending on their algorithmic features. NACK-based AODV (N-AODV) is a variant of the well-known Ad-hoc On-demand Distance Vector (AODV) reactive protocol for MANETs which we proposed with the aim of improving the NTA of the original protocol. In this paper, an experiment aimed at comparing N-AODV to AODV is reported; it moves from an exploratory case study we conducted preliminarily. The results obtained show that a MANET adopting N-AODV exploits higher NTA than a MANET adopting AODV. Moreover, the improved awareness positively impacts effectiveness.

Abstract State Machines (ASMs) have been successfully applied for modeling critical and complex systems in a wide range of application domains. However, unlike other well-known formalisms, e.g. Petri nets, ASMs lack inherent, domain-independent characterisations of computationally important properties. Here, we provide an ASM-based characterisation of the starvation-free property. The classic, informal notion of starvation, usually provided in literature, is analysed and expressed as a necessary condition in terms of ASMs. Thus, we enrich the ASM framework with the notion of vulnerable rule as a practical tool for analysing starvation issues in an operational fashion.

"Avanzi 3-Grano 23" (G23) is an old variety of Triticum aestivum from the mountain areas of Lunigiana (north Tuscany, Italy), where traditional farming communities have contributed to its success and on-farm conservation. G23 flour, traditionally used for typical food products, is characterized by particular nutritional and sensory traits but has technological properties which limit its suitability for breadmaking. The aim of this work was to evaluate how to promote the use of G23 through the optimization of bread formulation by leveraging both flour blending and the leavening system. During the preliminary test, three different mixes of G23 flour and a strong flour (C) were tested in terms of their leavening power as a function of leavening agent (baker's yeast or sourdough). The selected M2 flour, composed of G23:C (1:1 w/w), was used for further breadmaking trials and 100% C flour was utilized as a control. The sourdough bread obtained with the M2 flour (SB-M2) showed an improved sensory profile compared with the related control (SB-C). Furthermore, SB-M2 exhibited the best aromatic (high content in aldehydes, pyrazines and carboxylic acids) and phytochemical profile (total polyphenols and flavonoids content and antioxidant activity). In contrast, the use of baker's yeast, although optimal from the point of view of breadmaking, did not result in the same levels of aromatic complexity because it tends to standardize the product without valorizing the sensory and nutritional qualities of the flour. In conclusion, in the experimental conditions adopted, this old wheat variety appears to be suitable for the production of sourdough bakery products.

Primary murine mammary epithelial cells cultured on a laminin-rich-extracellular matrix (ECM) require c-Jun N-terminal kinase (JNK) activity for acinus formation. Inhibition of JNK (using SP600125) or small interfering RNA-mediated knockdown of JNK1 blocked acinus formation, impaired cell polarisation and lumen clearance and allowed sustained extracellular signal-regulated kinase (ERK) phosphorylation, cell proliferation, adhesion-independent cell survival and expression of epithelial-mesenchymal transition markers. ERK inhibition abolished the effects of JNK blockade. Interestingly, inhibition of JNK from the time of cell seeding blocked cell polarisation and lumen clearance; later inhibition (> 6 h) only affected lumen clearance. ERK inhibition effectively protected cell polarisation but less so, lumen clearance. SP600125-treatment similarly affected acinus formation by the ‘normal’ human mammary epithelial MCF10A cell line. Expression of dominant-negative JNK1 in MCF10A cells also undermined acinus formation, generating large ‘multi-acinar spheres’ whose formation is probably driven by excessive luminal cell proliferation and cell survival. As JNK activity must be suppressed from the time of cell seeding to block cell polarisation, we studied the behaviour of MCF10A cells immediately after seeding in laminin rich matrix: we detected engagement of cells with the matrix, early polarisation, movement of cells into clusters and ‘epithelial-cell- like’ behaviour of clustered cells. Inhibition of JNK activity or expression of dominant-negative JNK1 allowed cell engagement to the matrix, but blocked cell polarisation and all subsequent ‘behaviours’. While integrin activation occurred, tyrosine-phosphorylation of paxillin, Fak and Src was significantly damped by JNK inhibition. These results emphasise the multi-phase dependency of the organisation of mammary cells in 3D on JNK activity and suggest a ‘permissive’ support of ECM-integrin ‘outside-in’ signalling and a ‘damping’ of growth-factor ERK signalling as its two key cell physiological effects.

The shelf-life of bread is influenced by flour components, such as starch, composed of amylose and amylopectin. The aim was to test the effect of different balances of N (45, 90, 135 kg/ha) and P (48, 96 kg/ha) fertilizers on the flour characteristics and consequently the shelf-life of PDO Tuscan bread, stored in different modified atmosphere packaging (Ar, N2, Air). The amylose and phytochemical compounds were increased by N and decreased by the addition of P, but excessive doses of N (135 kg/ha) had a negative effect on flour quality. In the bread, the study highlighted the tendency of N2 and Ar, as storage filler gases, to reduce water loss, slow down the staling process, and prolong shelf-life. However, the most significant influence on shelf-life was related to the different fertilizations of wheat. In fact, when N was present in equal dose to P (90/96 or 45/48 kg/ha) or slightly higher (90/48 kg/ha), the bread tended to last longer over time. Instead, when these ratios were unbalanced in favor of N (135/48 or 135/96 kg/ha) and in favor of P (45/96 kg/ha), the shelf-life decreased considerably.

Lead poisoning from spent ammunition is known to affect many avian species. Birds of prey ingest lead when feeding on game shot with lead gunshot or bullets. Raptors with scavenging habits are particularly vulnerable to ingesting lead in areas with intensive hunting and are good indicators of the risk of poisoning from lead ammunition. To assess how much facultative and obligate avian scavengers suffer lead contamination in south-central Europe, between 2005 and 2019 we collected and analysed 595 tissue samples from 252 carcasses of 4 species (golden eagle, bearded vulture, griffon vulture, cinereous vulture). Lead concentrations in organs showed a similar pattern across species with long and small bones revealing the highest median values (5.56 and 6.8 mg/kg w.w., respectively), the brain the lowest (0.12), and the liver and kidney the intermediate (0.47 and 0.284). Overall, 111 individuals (44.0%) had lead concentrations above background thresholds in at least one tissue (i.e. >2 mg/kg w.w. in soft tissues, >8.33 in bone) and 66 (26.2%) had values indicating clinical poisoning (>6 mg/kg w.w. in liver, >4 in kidney, >16.6 in bone). Tissue lead concentrations and incidence of clinical and sub-clinical poisoning were higher in golden eagles and griffon vultures than in bearded and cinereous vultures, likely due to different feeding habits. In all species we found a rapid increase in lead values with age, but differences between age classes were significant only in the golden eagle. Birds with lead fragments in their digestive tract, as detected by X-rays, had higher median lead concentrations, suggesting that hunting ammunition is the main source of lead poisoning. Our results imply that lead impacts the demography of these long-lived species with delayed sexual maturity and low reproduction rate. A rapid transition towards lead-free bullets and gunshot is therefore required across Europe.

When bottled wine is opened, a completely different scenario occurs that can accelerate the oxidation of the product. This is called the secondary shelf life (SSL), which is generally shorter and less predictable than the primary shelf life (PSL). In this context, the research aim was to evaluate the changes that occur in two types of red wine during two tests to evaluate the secondary shelf life as a function of the packaging systems. The variation of Total SO2 and Free SO2 and the other chemical parameters (polyphenols, anthocyanins, proanthocyanidins, color, and volatile acidity) were used to assess the oxidation rate of the packaging samples after opening during the SSL. In both tests and for the two types of stored red wine, the polymeric cap showed the best results. The other types of closure (screw cap, natural cork, crow cap, and Tetra Brik) showed a negative trend and a reduced SSL for both red wines. Finally, the sensory results confirmed that with the polymeric cap, the SSL increases considerably compared to other capping systems. These results may be due to the technical characteristics of polymeric materials, which tend to vary slightly in shape after repeated usage.

Rap1p, the major telomere repeat binding protein in yeast, has been implicated in both de novo telomere formation and telomere length regulation. To characterize the role of Rap1p in these processes in more detail, we studied the generation of telomeres in vivo from linear DNA substrates containing defined arrays of Rap1p binding sites. Consistent with previous work, our results indicate that synthetic Rap1p binding sites within the internal half of a telomeric array are recognized as an integral part of the telomere complex in an orientation-independent manner that is largely insensitive to the precise spacing between adjacent sites. By extending the lengths of these constructs, we found that several different Rap1p site arrays could never be found at the very distal end of a telomere, even when correctly oriented. Instead, these synthetic arrays were always followed by a short (≈100-bp) “cap” of genuine TG repeat sequence, indicating a remarkably strict sequence requirement for an end-specific function(s) of the telomere. Despite this fact, even misoriented Rap1p site arrays promote telomere formation when they are placed at the distal end of a telomere-healing substrate, provided that at least a single correctly oriented site is present within the array. Surprisingly, these heterogeneous arrays of Rap1p binding sites generate telomeres through a RAD52-dependent fusion resolution reaction that results in an inversion of the original array. Our results provide new insights into the nature of telomere end capping and reveal one way by which recombination can resolve a defect in this process.
